First you should activate the DRAW tab from File/Options/CustomizeRibbon/DRAW

After that you can select Pen tool and set your thickness.
Your drawings will be vector, so you can set its thickness after you draw and you can replay the drawing by adding REPLAY animation.

It’s possible to create a Whiteboard animation this way.
